Osteosarcoma Market is Set to Rise by 2036 as 8 Emerging Therapies Set to Drive Future Growth

Osteosarcoma Market is Set to Rise by 2036 as 8 Emerging Therapies Set to Drive Future Growth

DANYELZA (naxitamab-gqgk) is a humanized anti-GD2 monoclonal antibody designed to recognize GD2-expressing tumor cells and promote antibody-dependent cellular cytotoxicity. Early investigations have reported manageable safety and encouraging activity, although additional evidence is required to establish its safety and effectiveness in older osteosarcoma populations.

SERB Pharmaceuticals acquired Y-mAbs Therapeutics in September 2025, adding DANYELZA to its rare oncology portfolio and strengthening its capabilities in targeted cancer therapeutics.

Sadaf Javed, Functional Head of Forecasting and Analytics at DelveInsight, stated that the asset carries a "selective responder" profile rather than broad efficacy, with commercial potential constrained by small eligible populations and uncertain tolerability in older osteosarcoma patients. Overall, it is viewed as a niche orphan oncology product with modest expansion upside rather than a high-growth oncology franchise.

Aminex Therapeutics' AMXT 1501 + DFMO

Polyamine uptake inhibitors

Aminex Therapeutics is developing AMXT 1501, an oral proprietary polyamine uptake inhibitor, in combination with DiFluoroMethylOrnithine (DFMO or eflornithine). The combination targets complementary components of polyamine metabolism and is intended to enhance immune-mediated antitumor activity.

The approach is being investigated as an immuno-oncology strategy across solid tumors, including osteosarcoma, and represents an alternative mechanism that could complement conventional anticancer treatments.

MedPacto's Vactosertib

TGF-β1 kinase inhibitor

Vactosertib (TEW-7197) is a TGF-β1 signaling inhibitor designed to modify the tumor microenvironment. Abnormal TGF-β activity can suppress immune responses, support metastatic processes, promote tumor-associated fibrosis, and contribute to resistance against anticancer therapies.

By interfering with these pathways, vactosertib could potentially improve immune-cell penetration and enhance the effectiveness of other treatments. The FDA granted Fast Track Designation to vactosertib in January 2023 for relapsed/refractory metastatic osteosarcoma. It previously received Rare Pediatric Disease Designation, while the European Commission granted Orphan Drug Designation in July 2023.

According to Javed, vactosertib is a high-upside but still unproven platform asset, with valuation and outlook hinging on whether later-stage studies can confirm consistent response rates and clinically meaningful durability in a broader population.

GSK's Risvutatug Rezetecan

ADC targeting B7-H3

Risvutatug rezetecan (Ris-Rez; GSK5764227; HS-20093) is an investigational ADC targeting B7-H3, a protein expressed across multiple solid tumors. The therapy is designed to bind B7-H3-positive cancer cells and deliver a cytotoxic payload directly to them, potentially improving tumor selectivity.

The candidate is being evaluated in GSK's global EMBOLD development program across several tumor types. In 2025, the FDA granted Breakthrough Therapy Designation to risvutatug rezetecan for patients with late-line relapsed or refractory osteosarcoma, highlighting its potential in a population with significant unmet medical needs.

Peel Therapeutics' PEEL-224

DNA topoisomerase I inhibitors

PEEL-224 is a polymer-based nanocarrier containing multiple small molecules and is being investigated as a potential treatment for aggressive cancers. Its active component is derived from camptothecin, a naturally occurring compound that targets topoisomerase I and disrupts DNA replication.

The polymer-based design aims to improve drug delivery while reducing toxicity and limiting drug efflux through the ABCG2 multidrug transporter. Preclinical research has reported sustained complete responses in certain aggressive solid-tumor models, suggesting potential activity against treatment-resistant disease.
